The effect of Aluminum ion in the SH-SY5Y cell - A preliminary proteomic study for Alzheimer’s disease. / 鋁離子對SH-SY5Y細胞之影響-應用於阿茲海默症之蛋白質體研究

碩士 / 高雄醫學大學 / 醫學檢驗生物技術學研究所 / 101 / Alzheimer’s Disease (AD) is a common elderly dementia. Over-in taking aluminum ion and prevailing AD was published highly relevant. The objective of this study was to simulate an AD environment by stimulating SH-SY5Y with aluminum ion. This research focused on association of oxygen stress in cells and AD after treating ions. Proteomics analyses were performed on the variation of protein in cell. Four proteins were identified, CBP80/20-dependent translation initiation factor (CTIF), Early endosome antigen 1(EEA1), Leucine-rich repeat neuronal protein 4 (LRRN4) and Phosphatidylinositol 3-kinase regulatory subunit beta(PI3KR2). Difference of those four proteins were monitored by Western blotting and IF. PI3KR2 inducing β-amyloid increase was discovered. The downstream product of PI3KR2, GSK3β, over-phospholated Tau protein. β-amyloid and Tau protein were factors of AD. As a result, aluminum ion may cause risk of AD.
