Studies in the Florfenicol to control the Bacterial Disease in the Sturgeon, Huso huso / 氟甲磺氯黴素在鱘龍魚之細菌性疾病防治研究

The aim of this study was to develop a technique to study the pharmacokinetics and determinate the residual of florfenicol(FFC) in sturgeon by high-performance liquid chromatography(HPLC)with UV detector.Using 0.05 M ammonium acetate(pH7.2)-acetonitrile-tetrahydrofuran(76:23:1)as the mobile phase at a flow rate of 1.0 ml/min, our developed HPLC method for the determination of FFCwas carried out on a C18 column(Cosmosil 5C18-MS, 150 mm X 4.6 mm I. D)The wavelength of UV detector was set at 223 nm. The residues of FFC in the muscle and liver of sturgeon extracted by solid-phase extraction method were detected with our developed method in this study. The treated tissue was cleaned by DDW and n-hexane then eluted by acetonitrile. The retention time of FFC was about 7.3 minutes. The limited detection concentration of our developed method was 0.01 mg/kg.After given single dose10 mg/kg-bw/day of FFC, via oral administration the pharmacokinetic parameters Cmax, Tmax, AUCinf , T1/2, Cl, V/F and MRT were 10.9568 μg/mL, 10 hr, 251.064μg.hr/mL, 12.1301 hr, 0.0387L/kg,0.6777 L/kg,23.7431hr, respectively. The results revealed that the recoveries of muscle and liver from sturgeon were 92-108% and 90-104%, respectively. The amount of the FFC in the fish muscle of the tenth daywas less than 1 ppm;however, the amount of the agent in the liver as compared as the seventh day. The allowed amount of FFC in the fish meat in Taiwan is less than 1 ppm;therefore, the recommended withdrawal period is 15 days according our study results.
