Extraction, Purification and Preservation of C-phycocyanin from Spirulina platensis Biomass / 螺旋藻之藻藍素的萃取、純化、及活性保存之研究

碩士 / 國立成功大學 / 化學工程學系碩博士班 / 101 / C-phycocyanin (C-PC) is water-soluble and can be easily extracted from Spirulina sp. as a protein-pigment complex. C-PC has been used in health drinks, food colorant and cosmetics. Furthermore, some of the medicinal properties such as antioxidant, anticancer activity are proven recently.
Here we developed a simple method for obtaining C-PC with high purity without the use of ion exchange chromatography. The procedure is based on enzymatic hydrolysis to dissolve outer cell wall of Spirulina sp. biomass, followed by solvent extraction to gain crude extract of C-PC with high initial purity, then purification by pigment precipitation through ammonium sulfate saturation, finally tangential flow filtration was implement to desalt and concentrate product.
The commercial application of C-phycocyanin is limited due to its sensitivity to heat, light, microorganism, etc, which results in precipitation and fading of the blue color, and C-PC will lose its biological activity. In this study, we have tried to use different methods to stabilize C-PC under room temperature, which included making C-PC into powder form and adding preservatives into C-PC solution. Finally, low dose of EDTA-2Na to C-PC (weight ratio 1:5) was found to be an effective preservative in pH6.0 C-PC solution, which can maintain its 90% initial activity after 35days under room temperature.
