The Bioactivity Study of Enzymatic Hydrolyzed Fish ScaleCollagen from Taiwan Tilapia / 酵素水解之台灣鯛魚鱗膠原蛋白生物活性之研究

碩士 / 國立成功大學 / 化學系碩博士班 / 101 / The fish scale was once thought of a kind of trash for Taiwan Tilapia, Right now, it has become a convient source of collagen. In this study, we focus on the extraction and hydrolysis of the scale collagen, then investigate the relationship between the bioactivity and the molecular distribution of hydrolyzed peptide. The cleaned scales were autoclaved for 60 min. The molecular weight distribution of the liquid portion was analyzed by using Superdex peptide® 10/300 column, and found that there were three distributions, 12500Da, 12500Da~6511D, and 6511Da~1355Da, centered at 6511Da~1355Da. The ability for scavenging free radical, reduction, and chelating with Cu2+ were 49.65%, 0.67(OD 700nm), and 69.89%, respectively. The autoclaved scales was incubated at 50℃ for 7 hours with the presence of Bromelain, papain, and Flavourzyme, respectively, it was found the major distribution of molecular weight of the hydrolyzed peptides was centered at 6511Da ~ 1355Da. And the bioactivity was found increased by about 60% for scavenging the free radical and 30% for reduction. With the increase of hydrolyzed time, the bioactivity increases, indicating that the peptides are shorter. The assay to test the curing effect of the cut wound of rat, it was found that there was no significant difference compared with the control one; however, it was observed that the hydrolyzed fish scale collagen has effect on stimulate the growth of hair around the cut wound.
