碩士 / 國立嘉義大學 / 食品科學系研究所 / 101 / This study is based on previous collection of the aquatic product tests between 2010 and 2012 in laboratory of Dr. Jian-Jeng, Huang at DFS, NCYU and Department of Health plan Taiwan, the results showed that Eleutheronema rhadinum, Trachinotus blochii, Rhabdosargus sarba, Meretrix lusoria, Aurigequula fasciatus, Lutjanus erythropterusare of high-risk drug residues aquatic species, while high-risk drugs include malachite green and leucomalachite green, furazolidone metabolite AOZ, sulfonamides and quinolones, these four drugs in fish should confined to food regulations of nontolerated/nondectectable levels.
At first stage, 40 high-risk species were sampled, these four different items were tested by different market-available brands of enzyme immunoassay test kits, and each of them were subjected to LC/MS/MS detection for comparison. The results showed two selected enzyme immunoassay reagent kits presented the same accuracies of up to 87.5% toward malachite green detection, while the probability of false negatives can be reduced to 6.06% and 8.57%. Four enzyme immunoassay reagent kits of furazolidone metabolite (AOZ) are of accuracies up to 100%. Two enzyme immunoassay reagent kits of sulfonamides detection present accuracies up to 90% and 97.5%; and both indicate the existence of other sulfonamide cross-reactivity, the two kits can be used as multiresidue analysis of sulfa drug residues, including sulfathiazole, sulfamethoxazole, sulfaquinoxaline, sulfadiazine. Two sets enzyme immunoassay reagent kits of quinolones detection possess accuracies up to 95% and 97.5%; and both accompany along with other quinolones cross-reactivity, it can be used as multiresidue analysis of quinolones residues, including norfloxacin and enrofloxacin.
At second stage of the experiments, 198 random aquatic products were sampled using an average sampling plan. The same four drugs residues enzyme immunoassay kits for rapid detection of all positive samples and randomly selected negative samples were re-confirmed by LC/MS/MS and for comparison study. The accuracies of malachite green and leucomalachite green enzyme immunoassay reagent kits, used previously and other than the selected kits in this study during 2010-2012 in the laboratory of Dr. Jian-Jeng, Huang, were 79.2%, 72% and 79.44%, respectively; and it should noted that the previously used kits caused serious false positive and false negative occurances. The results of this present study showed that the most out-prerformanced enzyme immunoassay kit from the first stage increased accurate rate to 91.67%, while the accuracies of furazolidone metabolite (AOZ), sulfonamides and quinolones were 100%, 92% and 91.67%, respectively. The screened enzyme immunoassay kits corresponding to the four designated items can cover eight out of ten high-risk drug residues, although the study results showed high accuracy of enzyme immunoassay kits, but there is still very small part of false positive and false negative occurrances. Nevertheless, according to the results of this study, these enzyme immunoassay reagent kits can effectively detect the eight high-risk drug residues, for rapid screening of aquatic products in efficient manners.
In this study, the cursory cost evaluation of the selected enzyme immunoassay reagent kits is expected to reduce about NT $ 12,525 per sample comparing to the current charge of liquid chromatography tandem mass spectrometry performed by Ultra Trace Industrial Safety Hygiene, and the enzyme immunoassay kit used for analysis for mass loading of aquaculture products in a considerably short time, which liquid chromatography tandem mass spectrometry would be rarely comparable. The results of this study found that the selected enzyme immunoassay kits used are of high accuracy. The sustained for rapid screening simulation test would be necessary for a scale-up sample size with the targeting accuracy higher as to establish the rapid screening method for aquatic products in Taiwan.
