Function Of Autocrine Motility Factor Receptor (amfr) In The Epibolic Movement Of Zebrafish Gastrula / 斑馬魚自分泌移動因子受器在胚胎原腸期外包過程中所扮演的角色

碩士 / 國立高雄海洋科技大學 / 海洋生物技術研究所 / 101 / Autocrine motility factor receptor (AMFR) is a membrane glycoprotein, also called gp78. AMF/GPI is its ligand. On the cytoplasmic membrane, AMF/GPI can bind AMFR and stimulate cell migration.When expressed on the membrane of endoplasmic reticulum, AMFR plays a role as E3 ubiquitin ligase that is involved in the endoplasmic reticulum associated degradation (ERAD). amfr is expressed in the yolk syncytial layer (YSL) from the blastulation to gastrulation during embryonic development. YSL can further be divided into internal YSL (i-YSL) and external YSL (e-YSL). YSL plays important roles in the embryonic development. Knockdown of amfr by amfr AUG morpholino produced defective phenotypes and caused epibolic delay in zebrafish embryos. The defective morphant phenotypes could be rescued by amfr capped mRNA. On the other hand, microinjection of amfr splicing morpholino caused amfr primary mRNA alternative splicing, but did not significantly cause epiboly delay. On the agarose gel, we found the larger alternation splicing fragment, called variant 1 (V1), and normal size of 291 bp. After sequencing, we found the smaller variant 2 (V2) fragment. In situ hybridization using sb:cb37(cb37) as YSN marker was used to observe embryonic YSL. We found the leading YSN of external-YSL were irregularly aligned. Using epcam as a marker for embryonic EVL, knockdown of amfr caused epibolic delay but the leading edge of EVL were normal. Thus, Knockdown of amfr expression caused epiboly delay, the leading edge of external-YSN were irregularly aligned.
