碩士 / 國立高雄海洋科技大學 / 水產食品科學研究所 / 102 / In this study, the effects of high temperature and high pressurepuffing parameters and two extraction methods (A and B) on the extraction mechanism and yield of fucoidan from Sargassum cristaefolium were investigated. Thin layer chromatography(TLC) and Fourier transform infrared spectrometer(FTIR) methods were used toidentify the characteristics of fucoidan and the antioxidant and antitumor activities of fucoidan were also examined. Results suggested thatproximate analyses for dried Sargassum cristaefolium were moisture(8.49%), ash(22.69%), crude fat(1.38%), crude protein(3.50%), and carbohydrates (63.93%). After high temperature and high pressure puffing treatment, the intensity of offensive smell and moisture content of algae were significantly decreased, the cell walls of pseudobulb and false vanes were obviously destroyed, and the bulk density and expansion ratio of algae were significantly increased. Moreover, the extraction yield of algae polysaccharide for method A is higher than that of method B. Elevated puffing temperature increased the extraction yield of algae
polysaccharide in both A and B extraction conditions. Through TLC and FTIR analyses, it was confirmed that algae polysaccharide possessed fucose and sulfate ester group. The algae polysaccharide from method B had better antioxidant activity than that of method A, and differnent puffing temperatures (140℃, 180℃, 220℃) affected the antioxidant activity of algae polysaccharide significantly. Algae polysaccharides from method A and B both showed antitumor activity.
