碩士 / 國立屏東科技大學 / 動物疫苗科技研究所 / 101 / Streptococcus iniae (S. iniae) has become an important aquatic pathogen for few decades and caused serious streptococcosis outbreaks of cultured fish in the world including Taiwan. However, there is no effective measure to manage the impacts generated from the streptococcal infections. It is very urgent to develop a useful and practical vaccine against streptococcal infections in cultured fish. In our laboratory, the proteomic technique has been used to study the protein profile of S. iniae and evaluate possible virulence factors. Two proteins, superoxide dismutase (SOD) and glyceraldehyde 3-phosphate dehydrogenase (GAPDH), have been considered as crucial virulence factors for bacterial infection. In this study, we have cloned SOD gene from S. iniae and produced the purified recombinant SOD (rSOD). The protein was then mixed with recombinant GAPDH (rGAPDH) that we have cloned previously in our laboratory. The protective efficacy of the mixed vaccine containing rGAPDH and rSOD was evaluated in tilapia. Fourteen days after priming, ELISA results indicated that the immunized groups except
inactivated vaccine group showed significantly higher antibody responses than those of PBS group. More importantly, antibody responses induced by inactivated vaccine +rSOD + rGAPDH were higher than those contributed by inactivated vaccine. Thus, rGAPDH and rSOD could induce the tilapia humoral immune response in a short time. In antibody responses from immunized fish in all groups except rSOD-immunized group were enhanced by comparison with immune responses between priming and boosting. Two weeks after boosting, all fish were challenged with 5.5x108 CFU of S. iniae. Four weeks after challenge, the relative percent of survival (RPS) for each group was calculated. RPS of the rSOD-immunized group was 84.66%, RPS of the rGAPDH-immunized group was 61.63% and RPS of the inactivated vaccine group was 100%. The group of fish immunized with inactivated vaccine +rSOD + rGAPDH-immunized showed 100% RPS. Based on the results, antigens selected by the proteomic technique provide partial protective capacities. Both groups immunized with inactivated vaccine or inactivated vaccine +rSOD + rGAPDH show strong protection against S. iniae infection in tilapia.
