碩士 / 國立臺灣海洋大學 / 食品科學系 / 101 / The aims of this research are to investigate the anti-inflammatory and anti-prostate cancer effects of Sargassum cristaefolium, a kind of brown seaweed, using cell models. The polysaccharides of S.cristaefolium inhibited nitric oxide (NO) production in the time-and concentration dependent manners in LPS-induced RAW264.7 macrophage, with 64.77% inhibition at 400 μg/mL. This polysaccharides was further proved to inhibit NO production through MAPK signal pathway.It could inhibit in a concentration-denpendent manner the phosphorylation of signal proteins of p-38, ERK and JNK and accordlingly, inhibit the activation of the transcription factor NF-B and the expression of inducible nitric oxide synthase (iNOS).Sargassum polysaccharide could only decrease the survival of LNCaP cell but not PC-3 and 22Rv1 prostate cancer cells. 400 μg/mL polysaccharide, cell surivival rate for LNCaP cells was 78%.The proposed mechanism for this polysaccharides on LNCaP cells was provided. This polysaccharide at 400 μg/mL enhanced ROS production from 19.2% to 47.3%, decreased mitochondrial membrane potential by 2.33 fold and let cell arrest in Sub G1 ans S phase.A eluate, Sc F80 from the ethanol extract of S.cristaefolium was demonstrated to decrease survival rate of PC-3 cells, with 45% survival rate at 50 μg/mL. Sc F80 induced intracellular ROS production in PC-3 in a concentration-dependent manner. At 50 μg/mL, it increased ROS production by 11 fold and decrease mitochondrial membrane potential by 17 fold. Sc F80 cause apoptosis of PC-3 cells and let cell cycle arrest in G2/M phase.
