Identification of Chlorella phytosterols / 小球藻植物固醇之鑑定

Phytosterols are essential components of cell wall and cell membranes for plant and algae. Their biological functions appear to maintain adequate function of plant cell membranes, control membrane fluidity and permeability, modulate the activity of cell membrane-bound enzymes such as ATPase and inhibit cholesterol absorption in animals. Phytosterol of Chlorella sorokiniana contained in stigmasterol, campesterol, ergosterol, brassicasterol and non-identified phytosterol. This research focused on the isolation and purification of non-polar compounds and phytosterols of Chlorella sorokiniana, the molecular structure of these compounds were also identified.
C. sorokiniana was extracted with dichloromethane and methanol (2:1, v / v). After celite solid phase extraction, the eluate underwent acid and base hydrolysis, and subsequently a series of column chromatography: diaion HP20, silica gel, silver-nitrate silica gel column chromatography. Finnaly, GC/MS and NMR were used to identify two major sterols, ergosterol and 7-dehydroporiferasterol. Moreover, phytol has also been isolated from C. sorokiniana during this isolation and purification. Phytol is produced by the basic hydrolysis of chlorophyll. The content of major phytosterols in C. sorokiniana, ergosterol and 7-dehydroporiferasterols were 1105.70, 1747.79 (g/g dry algae) restpectively, the content of brassicasterol, stigmasterol, beta-sitosterol, cholesterol and campesterol were 142.7, 165.90, 36.59, 18.58 and 7.71 (g/g dry algae).
