Composition of picoeukaryotic Chlorophyta in the coastal water off the northeastern Taiwan / 臺灣東北部沿岸海域超微真核綠藻種類組成之研究

碩士 / 國立臺灣海洋大學 / 海洋環境化學與生態研究所 / 101 / Photosynthetic picoeukaryotes are the fraction of phytoplankton composed of cells with size less than 2 or 3 μm and usually dominated by Chlorophyta in the world ocean. Until now, the taxonomic composition of picoeukaryotic Chlorophyta in subtropical water is still not well known, especially in northwestern Pacific Ocean. The main objective of our study is to study the taxonomic composition and variation in abundance of picoeukaryotic Chlorophyta in the coastal water off northeastern Taiwan. For qualitative study we used flow cytometry to sort out photosynthetic picoeukaryotes then cloned the sorted cells with 18S rRNA gene to examine composition of picoeukaryotic Chlorophyta. For quantitative study method we adopted tyramide signal amplification - fluorescent in situ hybridization (TSA-FISH) to measure abundances of picoeukaryotic Chlorophyta. Our experiment was duplicated monthly from July to December 2012.
The results from the clone libraries of 18S rRNA gene revealed that Micromonas clade 2 was the dominant genus. A novel Micromonas clade, hereafter the Keelung group, was defined here as a new clade that may be a species of subtropical northwestern Pacific Ocean. This study and other investigations revealed that clades 1 and 2 were worldwide distributed; clade 4 was only found between latitudes 20o - 40oN; and clade 3 occurred in waters north of 20oN. Therefore, each of the five clades of Micromonas has different range of geographic distribution. Based on TSA-FISH analysis, abundance of Micromonas in our study area varied between 2 cells mL-1and 2.5 × 103 cells mL-1. Because the sequences of Keelung group are mismatched with the probe of Micromonas (MICRO01), its abundance might be underestimated.
