Effect of The Addition of Commercial Desalted Deep Seawater on Polysaccharide Production by Agaricus Blazei in Liquid Culture and Its Assays of Anti-tumor Activity / 添加市售去鹽化深層海水對姬松茸液態發酵產生多醣體之影響及其抗腫瘤活性分析

碩士 / 國立臺東大學 / 理工學院綠色科技產業碩士專班 / 101 / The objectives of this study are to explore the effect on production of extracellular polysaccharides of Agaricus through submerged fermentation along with the addition of commercial desalted deep seawater (CDDSW) and to analyze the antitumor activity of its extracellular polysaccharide product. After addition of 0 (the control group, i.e. without adding any CDDSW), 1, 10, 50, and 100% CDDSW in such cultivation of Agaricus for 21 days, its dry mycelium obtained turned out to be 5.9±0.4, 5.2±1.5, 5.7±1.9, 6.1±0 6 and 6.3±0.4 mg/ml, respectively. The extracellular polysaccharides produced after 7 days of cultivation was 20.7±8.2, 20.7±11.9, 22.5±7.0, 27.0±11.1, and 26.1±4.1 ug/ml, respectively in each of the above cases.
To understand the effect of Agaricus polysaccharides on the regeneration rate of repair cells and the cell regeneration by immune function, we used the Agaricus polysaccharide powder solution to feed some 4-week old ICR mice, whose spleens were 50% removed, and then evaluated its antibody titer produced after immunization with attenuated cobra venom.The results show 80% of the mice were antibody production positive, their body weights reached 21-27 gm, which means all of them were in normal growth range. Further analysis of antitumor activity of the polysaccharides was performed. The tumor growth time of the nude mice fed with the polysaccharides delayed 7-14 days compared to the control group, which were injected with lung cancer cells only. Through non-invasive in vivo imaging system (IVIS Spectrum Imaging System), no positive images rendered. No recurrence of tumor were observed in 29% of the experimental mice group, the result could be somewhat associated with the feeding of Agaricus.
