Effect of Astaxanthin and Collagen Peptide Complex on Anti-aging in Senescence Accelerated Mice / 探討蝦紅素與膠原蛋白胜肽複合物對老化促進小鼠延緩衰老之影響

碩士 / 靜宜大學 / 食品營養學系 / 101 / Previous studies suggest that astaxanthin and collagen all have antioxidant and neuroprotective function, but its composite effect is unclear on anti-aging. The purpose of this study was to examine the effects of astaxanthin and collagen peptide complex on anti-ageing in senescence accelerated mice (SAMP8). 3-month old male and female SAMP8 mice were divided into control and four experimental groups. Experimental groups were supplemented with astaxanthin and collagen peptide complex 2.9g/kg BW (0.5x dose group), 23.94 g/kg (1x dose group), 47.88 g/kg (2x dose group) and 71.82 g/kg (3x dose group) for 12 weeks. Each group contained 10 mice. Food intake and body weights were recorded every week. Open field test, grading score, single-trail passive avoidance test and active shuttle avoidance test were estimated in 11th and 12th week. The serum biochemical parameters and antioxidant capacity were analyzed after sacrificed. The lipid peroxidation of malondiadehyde (MDA) and activity of antioxidant enzyme including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GSH-Rd) and Catalase activities of liver in mice were examined. In addition, hippocampus and cortex inducible nitric oxide synthase (iNOS) level were assessed to study the effect of RBC on brain oxidative damage and using immunohistochemical staining to observe the brain β-amyloid protein deposition conditions. The results showed that food intake, weight changes and open field test among the groups were no significant. The experimental groups of SAMP8 mice compared with the control group can significantly increase the skin glossiness level, reduce coarseness of skin and total aging score (p<0.05). Learning and memory ability of experimental groups were better than the control group (p<0.05). The liver and blood analysis results show that experimental groups could significantly decreased the blood cell and liver of MDA content, upgrade activity of SOD, GSH-Px, GSH-Rd and Catalase (p<0.05). In addition, antioxidant enzyme activity of GSH in blood cell was significantly higher than the control group (p<0.05). Astaxanthin and collagen peptide complex could also improve amyloid protein deposition conditions of brain.
In summary, the supplement of astaxanthin and collagen peptide complex could enhance the antioxidant capacity, and then reduce brain oxidative state, thereby improving learning and memory ability to achieve the anti-aging effect.
