碩士 / 大同大學 / 生物工程學系(所) / 101 / Davallia mariesii Moore ex Bak. is a medicinal, perennial fern. A rapid propagation protocol of Davallia mariesii has been established in this study. For callus induction, 0.1 g prothallus explants were cultured in 1/2 MS liquid medium supplemented with 2 % sucrose and subcultured in the same medium every month. Numbers of green callus was proliferated and the best growth ratio was 2.5 times after 2 months. Globular callus was formed from green callus in a diameter around 0.15 cm subcultured in the same medium every 2 weeks for 3 months. For sporophyte induction, globular callus in an average diameter of about 0.21 cm were cultured in 1/2 MS liquid medium supplemented with 2 % sucrose and 1 mg l-1 2ip (2-isopentenyl adenine). Sporophytes were regenerated and the percentage of induction rate was 100% after 2 months. The average number of regenerated sporophytes was about 35 per globular callus. The survival rate of sporophytes was about 70 % after acclimatization for 45 days. It is estimated that 1 g of prothallus explants as a starting inoculum can propagate more than 22,280 plants after one year. Regenerated sporophytes were diploid after chromosome examination.
