The Effects of Anisomeles Indica、Eleutherococcus Senticosus and Cassia Tora Extracts on Activated T cell-Mediated Cytokines / 在活化的T 細胞中探討魚針草、刺五加及決明子的萃取物對細胞激素調節之影響

碩士 / 朝陽科技大學 / 應用化學系生化科技碩博士班 / 102 / Activated T lymphocytes play a pivotal role in the pathogenesis of cell-mediated autoimmune diseases and chronic inflammatory disorders. T cell triggered by the activation of the T cell receptor/ cluster of differentiation 3
(CD3) complex and costimulatory receptor CD28 leads to the expression of cytokines such as interleukin-2 (IL-2), IL-4 and IL-13. Transcription factors nuclear factor of activated T cells (NFAT) and nuclear factor-kappa B (NF-κB)play essential roles in regulating the cytokines expressions. Many studies report that phorbol myristate acetate (PMA) and ionomycin can be used to simulate T cell activation. Anisomeles indica (AI) has anti-human
immunodeficiency virus (HIV) anti-Helicobacter pylori, free radical scavenging, cyclooxygenase I inhibitory, anti-histamine, analgesic and anti-hyperalgesic activities. Eleutherococcus senticosus (ES) has anti-inflammatory, antioxidant, antiviral activity, immune protection and immune regulation. Cassia tora (CT) is associated with lowering blood pressure, inhibiting histamine release , anti-platelet aggregation, anti-fungal,
anti-mutagenic, anti-oxidation, and protecting liver damage, night blindness,hypertension, hypercholesterolemia and constipation. However, the effect of these herbs on T cell immunity are still unclear. Therefore, this study
investigate the effects of AI, ES and CT extracts on T cell activation. We found that AI extract decreased NFAT and NF-κB transcriptional activities.Furthermore, it inhibited the mRNA expressions of IL-2, IL-4 and IL-13 in
activated JK cells. ES and CT extracts could not repress the transcription factor activities. ES extract also did not decrease the cytokines expressions in activated JK cell. However, CT extract promoted the cytokines expressions in activated JK cell. Based on the above data, we suggest that AI inhibits IL-2,IL-4 and IL-13 mRNA expressions by reducing the activity of NFAT and NF-κB to provide the immunosuppressive activity. ES has not the inhibited effect in activated T cells. It has the immunosuppressive activity, possibly by inhibiting other immune cell actions. CT could increase the cytokines expression to achieve the immune stimulatory effect on T cells.
