碩士 / 大葉大學 / 環境工程學系碩士班 / 102 / Sodium bicarbonate was used as the carbon source for cultures of Nannochloropsis sp. in this study to investigate effects of light distribution, concentrations of sodium bicarbonate, concentrations of sodium nitrate and culture pH on growth of Nannochloropsis sp. and lipid contents in biomass. The temperature during the experiment was kept at 30 °C and salinity was maintained at 25 g/L. Experiments were conducted by one-factor-at-a-time method. The concentrations of sodium bicarbonate were 0.1, 0.2, 0.3 M, concentrations of sodium nitrate 0.1, 0.2, 0.3 g/L and culture pH 8.4 , 9, 9.5, 10. The highest specific growth rate was obtained under the condition with sodium bicarbonate concentration of 0.1 M, sodium nitrate concentration of 0.3 g/L and the for growth of Nannochloropsis sp. was culture medium is setting concentration by sodium bicarbonate 0.1, 0.2, 0.3 M, and the concentration of nitrate 0.1, 0.2, 0.3 g/L, and pH 8.4. The lipid content in biomass of Nannochloropsis sp. can be increased by a two-stage cultivation. The lipid content in biomass was 15.1% with the initial sodium nitrate concentration of 0.5 g/L and sodium bicarbonate concentration of 24 g/L after seven-day cultivarion. The lipid content in biomass was up to 23% in the consecutive cultivation.
The light intensities were measured in the phtobioreactor with optical densities in the range from 0.79 to 3.4 for cultures of Nannochloropsis sp.. The relationship between light intensity, optical density and light path obeys Lambert-Beer.s law. This relationship for cultures of Nannochloropsis sp. can be used to predict the light intensity distribution in phtobioreactors.
