The feasibility study of freshwater algae applied for determination of biological acute toxicity / 淡水藻類應用於生物急毒性檢測之可行性

碩士 / 弘光科技大學 / 環境工程研究所 / 102 / The primary objectives of this study are to investigate the acute biotoxicity indicators using algae assay procedure bottle test method. The measuring algae and cyanobacteria including Chlorella, Microcystin, Anabeana, Oscillatoria, Gloeotrichia, and Nostoc. The test toxins were copper sulfate (CuSO4), cadmium chloride (CdCl2) and sodium dodecyl sulfate (SDS), which were prepared in raw water. The chlorophyll-a, cells number (cell density) and cell viability as toxicity parameters. Stress produced by the toxins addition was analyzed by flow cytometry. Experimental results show that the detection of a variety of algae chlorophyll a half inhibitory effective concentration (Effective Concentration 50%, referred to as EC50) of EC50 and algal density indicators show the differences between the sexual considerable, mainly because of the accumulation of the algal cells amount of poison may interfere with the test measurement of chlorophyll a, resulting in higher EC50 calculated error. In addition, the use of algae and algal cell counts stained with and the number of dead cells by flow cytometry in vivo measurements of the statistical results of the EC50, EC50 and staining of cell density count is closer.
