A study on the nutrient requirements of the culture ofTetraselmis chui, Nannochloropsis oculata and Isochrysis galbana / 探討培養周氏扁藻、擬球藻和等鞭金藻營養鹽需求

Microalgae are widely used as a live feed for the larvae of marine fish and bivalve. Walne medium is usually used a nutrient source of microalgae in the laboratory. However, application of the Walne media in large-scale culture is limited by economic costs. Although agricultural fertilizer could be applied to large-scale microalgal culture, the microalgal density is too low to provide sufficient feed in aquaculture. Therefore, to develop a nutrient composition for large-scale culture of microalgae is an important issue. This study investigated that the effect of different percentage of Walne medium on the growth of Tetraselmis chui, Nannochloropsis oculata and Isochrysis galbana in the presence of major nutrients of 16 ppm NO3--N, 4 ppm PO43--P, and 3 mM Tris. It was found that there is no significantly different between the growth of T. chui cultured in 1 % Walne medium and that cultured in 100 % Walne medium. However, the growth of N. oculata cultured in 5 % Walne medium and I. galbana cultured in 7 % Walne medium is significant lower than they cultured in 100 % Walne medium. In experiment of the effect of EDTA on microalgal growth, T. chui and N. oculata does not need extra-additition of EDTA, but 18.2 μM EDTA is required for the growth I. galbana cultured in 7 % Walne medium. In experiment of the effect of Fe3+ on microalgal growth, optimal concentration of Fe3+ for the growth of T. chui, N. oculata and I. galbana are 0.48, 0.72 and 1.01 μM, respectively. In experiment of the effect of Mn2+ on microalgal growth, it is found that the addition of Mn2+ does not affect the growth of T. chui, N. oculata and I. galbana. Therefore, the new nutrient formula for a large-scale culture of T. chui is 1 % Walne medium, 6.5 μM EDTA, 0.48 μM Fe3+, and 0.09 μM Mn2+. The new nutrient formula for a large-scale culture of N. oculata is 5 % Walne medium, 6.5 μM EDTA, 0.72 μM Fe3+ and 0.09 μM Mn2+. The new nutrient formula for a large-scale culture of I. galbana is 7 % Walne medium, 18.2 μM EDTA, 1.01 μM Fe3+ and 0.126 μM Mn2+. The comparison in the biochemical composition of microalgae cultured between in respective new nutrient formula and in Walne medium was made. Protein content of T. chui cultured in new nutrient formula is significantly lower that that cultured in Walne medium. Lipid content of N. oculata cultured in new nutrient formula is significantly higher than that cultured in Walne medium. There is no significant difference in the biochemical composition of I. galbana cultured between in new nutrient formula and in Walne medium. Therefore, Walne medium could be replaced by a new nutrient formula in the large-scale culture of microalgae.
