Using the ratio of blood cytochrome c1/GAPDH to evaluate the brain apoptosis on the ayu (Plecoglossus altivelis) aging model / 以香魚老化動物模式的血液cytochrome c1與GAPDH之比值做為評估腦部細胞凋亡的指標

碩士 / 國立宜蘭大學 / 生物技術與動物科學系生物技術碩士班 / 102 / In the recent reports, significant increase and inactivation of GAPDH in brain were highly associated with the neurodegenerative disorders, such as Alzheimer's disease (AD) and Parkinson's disease (PD). As for cyto.c1, when the integrity of mitochondrial outer membrane is lost, the membrane-bound cytochrome c and then cyto.c1 release into cytosol or out of cell membrane. When a lot of cyto.c1 is able to be tested, it has a positive correlation to the apoptosis indicators of caspase 9 and caspase 3.
By the way, we assumed that ayu was just triggering inflammation in brain during spawning. It seems to be necessary to induce the protection from the higher level of GAPDH. And after spawning, the apoptosis increased by the high expressions of caspase 9 (200 ± 57%) and caspase 3 (180 ± 55%), and cyto.c1 proteins in brain (142 ± 43%). Also, these phenomena of GAPDH and cyt.c1 in brain were also expressed in the blood plasma. Therefore, we expected to use the ratio of cyt.c1/GAPDH in blood serum to evaluate the brain degeneration.
The results showed that during and after spawning of brain, compared to before spawning, the protein expression of GAPDH in brain was 111 ± 7% and 97 ± 20%, respectively (P<0.01). On contrast, the cytosol protein expression of cyto.c1 in brain was 72 ± 9% (P <0.001) and 142 ± 43%, respectively (P <0.01). The ratio of cyto.c1/GAPDH was 0.65 ± 0.08 during spawning and 1.33 ± 0.51 after spawning. In blood plasma, the ratio of cyto.c1/GAPDH was 0.44 ± 0.43 during spawning and 1.60 ± 0.68 after spawning.
We suggested that the ratio in blood less than 1 might mean the inflammation in brain to trigger apoptosis. And the ratio more than 1 might mean the brain apoptosis already. The ratio of cyt.c1/GAPDH in blood plasma would be a potentially biological marker for brain degeneration.
