Highly-sensitive cholesterol biosensor based on anemone-like ZnO nanostructures manufactured by a modified hydrothermal process / 利用修改之水熱法合成出海葵狀氧化鋅奈米結構應用於高敏感度的膽固醇生物感測器

碩士 / 國立臺南大學 / 電機工程學系碩博士班 / 102 / The anemone-like ZnO nanostructures have been synthesized by a modified hydrothermal method and were further adsorbed immobilized cholesterol oxidase (ChOx) as a nano-biosensor.
Firstly, the anemone-like ZnO nanostructures have been synthesized through adding a simple step in hydrothermal method, the step was preheated aqueous solution of zinc nitrate and HMTA aqueous solution respectively before the hydrothermal reaction began. The X-ray Diffraction patterns showed that the (002) plane of anemone-like ZnO was more prouded than other crystal plane. It exhibited the ZnO nanocrystal structures have c-axis preferred orientation. The anemone-like ZnO nanostructures were mainly influenced by preheating HTMA solutions from the evidence of photoluminenscent spectra. The best preheating temperature is 90oC and the best growth time is 9 hours for the nanostructures through HR-SEM observations. The anemone-like ZnO nanostructures with length of 2.10μm were well prepared. The cholesterol oxidase (ChOx) were immobilized on the anemone-like ZnO nanostructures as a cholesterol nano-biosensor. The effect of thickness of ZnO seed layer on the biosensor was also studied. Better sensitivity was found with thinner ZnO seed layers for growing anemone-like ZnO nano-biosensors. A bioelectrode (ChOx/ZnO/ITO/glass) with high sensitivity of 57.533µAmM-1 cm-2(1.488 µA (mg/dl) -1 cm-2), a wide sensitive range from 25 to 500 mg/dl and a low value of Michaelis-Menten constant of 3.64 mM were obtained by electrochemical measurements. With thinner ZnO seed layer, the response time is reduced from about25 seconds to less than20 seconds.
It is concluded that the anemone-like ZnO nanostructure is suitable for cholesterol biosensors with good performance. The thickness of ZnO seed layer will significantly affect the performance of the biosensor.
