Growth、activation of innate immunity and immune response of white shrimp Litopenaeus vannamei following long-term culture at different pH levels / 白蝦長期養殖於不同pH環境其成長、先天性免疫啟動與免疫因子

White shrimp juveniles (0.6 ± 0.05 g) which had been reared at different pH leavel (pH 6.8、8.1、9.2) for 24 weeks were examined for the survival, growth, immune activation, immune parameter, resistance against Vibro alginolyticus and expressions of lipopolysacchairde-β-glucan binding protein (LGBP), C-type lectin, peroxinectin (PX), integrin β, serine proteinase, prophenoloxidase (proPO)Ⅰ, proPOⅡ, α2-macroglobulin (α2-M), cytosolic manganese superoxide dismutase (cytMnSOD), mitochondrial manganese superoxide dismutase (mtMnSOD), extracellular copper-zinc superoxide dismutase (ecCuZnSOD), glutathione peroxidase (GPx), catalase, lysozyme, penaeidin 3a, transglutaminase (TG)Ⅰ, TGⅡ, heat shock protein 70 (HSP70). No significant difference in survival rate(80-83%)of shrimp was found among shrimp reared at three pH levels. Weight gain percentage was 1648 %, 1575 %, 1421 %, and length increase percentage of 158 %, 155 %, 150 % for the shrimp reard at pH 6.8, 8.1 and 9.2, respectively. Shrimp reared at pH 9.2 grew significantly slower. Activation of immunity and immune parameter of shrimp reared at pH 8.1 were higher than those of shrimp reared at pH 6.8 and 9.2. Shrimp reared at pH 9.2 showed higher expressions of immune-related proteins including LGBP, C-type lectin, PX, integrin β, proPOⅡ, cytMnSOD, mtMnSOD, ecCuZnSOD, GPx, catalase, lysozyme, penaeidin 3a, TGSⅠ, TGSⅡ, HSP70. In another experiment, shrimp which reared at different pH levels for 24 weeks were challenged with V. alginolyticus. At 24-144 h, cumulative mortalities of shrimp reared at pH 8.1 were significantly higher than those of shrimp reared at pH 6.8 and pH 9.2. Phagocytic activity and clearance efficiency of shrimp to V. alginolyticus were much lower in shrimp that reared at pH 9.2. In conclusion, shrimp reared at pH 8.1 exhibited higher activation innate immunity and immune parameters, and increased resistance against V. alginolyticus. Shrimp reared at pH 9.2, exhibited reductions in immune parameters, but showed higher expressions of immune-related proteins indicating modulation of innate immune system to prevent further perturbations.
