Study on the cultivation, seed stock and the phycobiliprotein crude extraction of two red macroalgae Grateloupia sparsa and Hypnea charoides / 兩種紅藻稀毛蜈蚣藻與長枝沙菜的培養,種子化及其藻膽蛋白粗萃之研究

Main purpose is to investigate the cultivation condition and method of filaments and disc from two kinds of red algaes. Use the filaments and disc for seed stock , And then try to suspension culture the filaments and thalli. Discussion the phycobiliprotein crude extraction of two kinds of red algaes’s filaments and each stages of the thalli. At the cultivation part, in the shading experiment covered area 80% is the better way to cultured filaments and disc. Although growth quantity less than 60%, less susceptible to pollute. In the light quality experiment, at the part of G. sparsa filaments, yellow light resulted in the highest production of filaments biomass is 454.00 ± 110.01 (N/E), red light resulted in the lowest production of filaments biomass is 174.00 ± 46.36 (N/E). At the part of G. sparsa disc ,blue light resulted in the highest production of filaments biomass is 36.00 ± 5.57 (N/E), red light resulted in the lowest production of filaments biomass is 18.00 ± 3.00 (N/E) ; At the part of H. charoides filaments, green light resulted in the highest production of filaments biomass is 433.33 ± 14.05 (N/E), red light resulted in the lowest production of filaments biomass is 137.00 ± 16.52 (N/E). At the part of H. charoides disc, green light resulted in the highest production of filaments biomass is 115.00 ± 8.89 (N/E), the rest were no statistically significant differences were steady growth. In the expanding experiment of use the 1g filaments after twelve weeks of cultivation, at the part of G. sparsa filaments, chopped filaments resulted in the highest production of filaments biomass is 2.5 g, no chopped filaments resulted in the highest production of filaments biomass is 1.7 g. At the part of H. charoides filaments, chopped filaments resulted in the highest production of filaments biomass is 2.2 g, no chopped filaments resulted in the highest production of filaments biomass is 2.7 g. In the expanding experiment of use the disc after fire months of cultivation, two kinds of red algaes’s disc are can use to expanding cultivation. Two kinds of red alglae’s filaments and disc after immobilized, and after four times test for one month of cultivation, at the part of Sodium alginate no matter the filaments and disc’s activity are better PVA- Sodium alginate. In the seedling experiment, at the part of material is cultivation of nylon rope for 30 days the average number of seedlings is 7.92 ± 1.24 (N/cm) more than cultivation of cotton rope for 30 days the average number of seedlings is 2.00 ± 0.51 (N/cm). At the part of seed is filaments for 30 days the average number of seedlings is 16.72 ± 1.34 (N/cm) more than cultivation of disc for 30 days the average number of seedlings is 4.32 ± 0.52 (N/cm). In the suspension culture, the small thalli in terms of 24 L: 0 D photoperiod culture can stimulate its growth rate. While the large thalli in the outdoor cultivation is the lower the temperature can faster the growth rate, as if cold weather after is the best season for cultivation. So winter is the best time to outdoor cultivation. At the extraction part, when phycobiliprotein precipitation the G. sparsa extract was added to a concentration of 80% ammonium sulfate and the H. charoides extract was added ammonium sulfate to a concentration of 60%, could have a best extraction respectively. The crude extract showed that the highest concentration of extract thalli is G. sparsa’s small thalli in 24 L: 0 D photoperiod under suspension culture at 20℃ after eight weeks. And the highest concentration of filaments is G. sparsa’s spherical filaments group in 12 L: 12 D photoperiod under suspension culture at 20℃ after eight weeks. Both are use freeze-dried and then grinding the cells broken is the best extraction conditions, more in line with economic benefits, is not only save time, and the yield is more higher.
