Study on the cultivation, seed stock and fucoidan extracted factors of the Laminaria japonica / 海帶養殖與種子化及褐藻醣膠萃取條件之研究

The filamentous gametophyte and sporophyte were preserved in order to establish the best L. japonica culture and "seed stock" techniques. It is first to discover that the zoospores in the unilocular sporangium directly germinated into the initial gametophyte which bursted the unilocular sporangium and then they were released outside. The results of the release time of the initial gametophyte, in the control group (white light) at 18℃was a minimum of 5 days. And in various of light quality, at 12℃and 18℃, in red light groups were 14 days and 12 days, light intensity was 6.84 μmol m-2 s-1 and 6.68 μmol m-2 s-1, respectively. Those results indicated increasing temperature that helps shorten the release time, however the red light would delay the release time. In the culture of the initial gametophyte using different light quality resulted that at 18℃in the yellow light group (22.36 μmol m-2 s-1) had the fastest growing, the algal length was 52.5±5.0 μm, and at 12℃in red light group (6.84 μmol m-2 s-1) had the slowest growth, the algal length was 17.5±5.0 μm, indicated light intensities and temperatures were the most important effects on the gametophyte growth. However single light quality contribute to continue the growth of gametophyte. Separated female and male gametophytes that cultured at low temperature (12℃) and low light intensity (11.34 μmol m-2 s-1), had continued to proliferate of gametophyte. Higher temperature (18℃) had only to stimulate male gametophyte asexual growth. The results of L. japonica haptera tissue culture, showed that it could develop into three algal morphologies, when it at 18℃and 33.33 μmol m-2 s-1 of light intensity for 4 months, it remained to grow of haptera, but grew into blades from the callus of haptera, while haptera were immersed in growth hormones for 1 month that stimulate the epidermis layer of haptera to grow into filaments. In addition, in the biomass results of female and male gametophyte showed that they at 18℃could reach their maximum SGR after 2 weeks of culture, SGR of female one was 0.473 g/w and male gametophyte was 0.507 g/w. The results of the growth filaments from haptera showed it reached its maximum SGR in 18℃,was 2.138 g/w, after 1 week of culture. Results of sodium alginate and with PVA immobilization for "seed stock" showed that female and male gametophyte could not survival in the beads sodium alginate and with PVA, however the results showed that the filaments of haptera could be alive in the beads for "seed stock". Finally, to analyze and compare of L. japonica fucoidan extracted contents at different conditions, the results showed that the mainly effects on the fucoidan extraction were pH value and temperature, which significantly effected the contents of fucoidan of fucose、uronic acid and sulfuric roots. However, in HPLC determined 6 kinds of composition from fucoidan were glucuronic acid, glucose, rhamnose, fucose, galactose and mannose. In which, glucuronic acid content was the highest (98%) after acid hydrolysis
