Inhibition of Angiotensin I Converting Enzyme and Dipeptidyl Peptidase IV of Chlorella and Freshwater Clam Hydrolysates / 綠藻與蜆肉水解物對血管收縮素轉換酶及二肽基胜肽酶 IV 活性之抑制

碩士 / 國立臺灣海洋大學 / 食品科學系 / 102 / The pulverized chlorella and freshwater clam were used as materials in this study. Pulverized chlorella were hydrolyzed by cellulase (Cellulase AP3) and protease (Protease N or Prozyme 6) at 50℃ for 1-5 hours. The pulverized Chlorella were hydrolyzed by Protease N and Cellulase AP3, then fermented with lactic acid bacteria up to 40h to obtain chlorella Fermented hydrolysates. The freshwater clam were hydrolyzed by protease at 50℃ for 5 hours. The inhibitory effect of hydrolysates and Fermented hydrolysates on activity of DPP-IV (Dipepidy peptidase IV) were investigated. The pulverized chlorella were hydrolyzed by Cellulase AP3 and Protease N, their inhibition on DPP-IV with IC50 being 5.03 mg/mL, and it had higher inhibitory on DPP-IV with IC50 being 4.85-5.18 mg/mL in <10 KDa fraction. Chlorella Fermented hydrolysates on DPP-IV with IC50 were 7.17 mg/mL. Chlorella Fermented hydrolysates were separated into six fractions by size exclusion chromatography on a Sephadex G-25 gel with BPG column. The fraction FP5 (M.W.= 232-204 Da) of hydrolysate showed the better inhibition on DPP-IV with IC50 being 1.43 mg/ml.The fraction FP5 (M.W.= 232-204 Da) of hydrolysate showed the better inhibition on ACE (Angiotensin I converting enzyme) with IC50 being 28.7 μg/mL.
