Inhibitory effect assessment of carrageenan on Dengue virus infection via real-time monitoring of cellular oxygen consumption rates within a microfluidic device / 細胞耗氧量感測裝置應用於藻類萃取物作用於登革熱病毒感染之效能分析

碩士 / 國立臺灣海洋大學 / 機械與機電工程學系 / 102 / A microfluidic device combined with a light modulation system was developed to assess the inhibitory effect of carrageenan on Dengue virus (DENV) infection via real-time monitoring of cellular oxygen consumption rates (OCR). Monitoring cellular OCR, which can reflect cellular metabolic activity, enable us to monitor virus infection process in real time and rapidly determine the antiviral activity of the potential drugs/chemical compounds. The time variation of cellular OCR of the single cells in-situ infected by DENV with a different multiplicity of infection (M.O.I) was first successfully measured within a microfluidic device. Influence of time points of carrageenan treatment on DENV infection was then examined by real-time monitoring of cellular OCR for three group: cells were pre-treated with carrageenan and then infected with DENV as a pre-treatment group, carrageenan was added to cells simultaneously with DENV as a virucide group, and cells were pre-infected by DENV and then treated with carrageenan as a post-infection group. By monitoring cellular OCR, we can rapidly evaluate the inhibitory effect of carrageenan on DENV infection less than 12 hours showing that carrageenan has a strong and effective anti-DENV activity for three groups, in which a virucide group was observed to have a high inhibitory effect. Moreover, the prior treatment with carrageenan after DENV infection for the post-infection group or increasing the duration time of treatment can effectively increase anti-DENV activity. Our proposed platform provides the capability to perform time-course or dose-response measurements of cellular metabolic activity that is caused by virus infection or the antiviral activity of the potential compounds via monitoring of the OCR in rapid and real-time detection.
