Establishment of TALENT-mediated mutants of CPE and Nhlh2 genes in zebrafish(Danio rerio) / 建立TALEN引發斑馬魚CPE及Nhlh2基因的突變

碩士 / 國立臺灣海洋大學 / 生物科技研究所 / 102 / Biosynthesis of neuropeptides and peptide hormones need the proprotein convertase and CPE two enzyme systems. CPE cleaves the basic residues from C terminal basic amino acid (arginine, lysine) to generate active neuropeptides and peptide hormones (such as: insulin, Neuropeptide Y (NPY) ... etc.). Carboxypeptidase E (CPE) was found in the brain and neuroendocrine systems (endocrine pancreas, pituitary, adrenal gland). In an organism, food intake is subject to energy storage and energy metabolism. In the Leptin pathway, Leptin promote proopiomelanocortin (POMC) generation, CPE catalytic POMC into α-MSH, MC4R is the receptor of α-MSH , when α-MSH and MC4R combination will lead to the generation of TRH and then trigger downstream reactions, thereby enabling the organism to use energy and movement. Losing function of CPE will cause the organism procuring metabolic syndrome. In mammals, CPE knockout experiments in mice have the phenomenon of obesity. The body fat content of CPE KO mice is two times more than that in the wild mice. Also, activity and calories burned rate of CPE KO mice is relatively low. Muscle strength and coordination of CPE gene knockout mice were relatively weak, and blunt to stimuli together. CPE gene knockout can cause abnormal mammalian obesity, abnormal nervous system and abnormal endocrine system. Idereal aim to establish CPE KO zebrafish, and use this as an in vivo platform to study the physiological function of CPE in zebrafish. We expect CPE knockout zebrafish also cause obesity and abnormal endocrine.
