Application of Hydrophobic Interaction Chromatography in Purification Process of C-phycocyanin / 疏水性層析法應用於藻藍蛋白純化程序

碩士 / 明志科技大學 / 化學工程系生化工程碩士班 / 103 / In this work, stirred fluidized bed hydrophobic chromatography was employed to purify the C-phycoayanin (CPC) from disrupted Spirulina platensis. To investigate the adsorption and desorption kinetic and thermodynamic studies, the experiments were carried out in a small stirred tank system. The results were further applied to investigate the dynamic adsorption and elution behaviors in small scale packed bed (i.d. 16 mm) chromatography under the various operating conditions. Finally, the purification process was scaled up to stirred fluidized bed (i.d. 25 mm) chromatography to directly purify CPC from unclarified feedstock. The recovery yield and purification factor were used to compare the experiment results obtained from expanded bed and/or packed bed chromatography under the similar conditions.
The optimal operating conditions for adsorption of CPC were found to be 0.75 M (NH4)2SO4 at pH 7, disrupted algae 1%, and 37 ℃. Moreover, the optimal elution for CPC was 20 mM phosphate buffer at pH 7 and 37 ℃. For small packed bed chromatographic systems, the optimal adsorption conditions were 0.75 M (NH4)2SO4 at pH 7, disrupted algae at 5%, aspect ratio (H/D) at 1.56 and flow rate at 100 cm/h. There is no much difference for the elution efficiency between 100 and 300 cm/h. Hence, the liquid velocity of 200 cm/h was chosen in this work. For 10% disrupted algae in large scale stirred fluidized bed purification process, the CPC can be recovered in yield of 36.35% and purification factor of 5.60. This novel purification process was shown to be more successful as compared to the expanded bed (yield 30.52% and purification factor 3.73) and packed bed (yield 28.75% and purification factor 4.24) purification processes.
