Approach The Potential of Gender Relative Genes on Male Tilapia (Oreochromis niloticus) Juvenile as a Biomarker for EDCs Like Measurement / 探討雄性吳郭魚幼魚性別相關基因作為環境荷爾蒙類似物檢測的潛力

碩士 / 國立嘉義大學 / 水生生物科學系研究所 / 103 / In this study, we were to detect of environmental hormone by the only male of tilapia (Oreochromis niloticus), an important species for Taiwan aquaculture. We treated the juvenile of tilapia in 17β-estradiol (E2) and 17α-methyltestosterone (MT), and analyze the mRNA expression of dmrt1, cyp19α and vtg by Q-PCR. The dmrt1 mRNA of gonad was significantly reduced by E2 at 7.2 μM, but has no significant difference of the control group. They have dose dependent of vtg and cyp19α with E2 at different concentrations. The MT reduced cyp19α mRNA, but induced vtg mRNA. In addition, we detect vtg performance at different concentrations for 7 days by the Bisphenol A (BPA) of environmental hormones and heavy metals cadmium (cd). The liver exposed cadmium, which determined to E2 contents by ELISA. The result was up-regulation of vtg mRNA by BPA at 0.438 μM and it has does dependent in different concentrations. Vtg gene expression is increased significantly and up-regulation E2 levels in the liver with exposure to 2.865μM Cd for 7 day.
Depend on the results that we suggested that the dmrt1, cyp19α and vtg can detect of environmental hormone. The vtg is the most sensitive than other gene. Adverse effect on reproduction for Male of tilapia, when disrupt hormone homeostasis in cadmium pollution to water.
