Cell-penetrating peptides (CPPs) are short peptides of protein fragment which are able to penetrate cell membranes into cells. Lipsome is one of the important drug carriers which is composed by amphiphile molecules. Moreover, liposome possesses well biocompatibility and biodegradability. In this thesis, the pentapeptide KTTKS, a cell-penetrating peptide is used as the hydrophilic end. Norcantharidin (NCTD), an anticancer drug , or rhodamine, a fluorescent material, is used as hydrophobic ends. Therefore, the bifunctional monomers of NCTD-KTTKS and rhodamine-KTTKS to prepare liposomes. SDS-PAGE analysis is a simple, rapid and important technology of studying unknow compounds in organism protein expression. SDS-PAGE is used to analyze protein expression in E. coli and zebrafish by functional liposomes composed of cell-penetrating peptides. The results showed that liposomes composed of rhodamine-KTTKS increases the growth inhibition of E. coli. Nine proteins were changed and three of those nine proteins were identified as glycine dehydrogenase (decarboxylating), elongation factor Tu 1, and glutamate/aspartate periplasmic-binding protein. Analysis of the zebrafish protein with the same strategy. Showed both liposomes of NCTD-KTTKS and rhodamine-KTTKS influence the amount of proteins expression in zebrafish. Moreover, it was found that 200 μM NCTD-KTTKS in liposomal form can inhibit the growth of zebrafish. Fluorescent optical microscopic studies reveals that rhodamine-KTTKS liposomes exhibit specific binding with kidneys and ganglia in zebrafish and therefore can be developed a contrast agent.
