Continued Investigation on Chemical Constituents and Biological Activities from a Formosan Soft Coral Klyxum flaccidum / 台灣產軟珊瑚Klyxum flaccidum化學成分及生物活性之持續研究

碩士 / 國立中山大學 / 海洋生物科技暨資源學系研究所 / 103 / Investigation on the chemical constituents of the acetone organic extract from the Formosan soft coral Klyxum flaccidum, collected from the Liuchiu Island. The study had isolated and separated seven new natural products, include two cembrane-type diterpenoids, flaccidenes A and B (1 and 2) and five steroids, klyflaccisteroids E (3), G–J (4–7) along with four known compounds 8–11. The structures of compounds 1–11 were established by spectral data analysis (IR, MS, 1D &; 2D NMR) and comparison of spectral data with those of the related known compounds. Compounds 1 and 2 are unprecedented a new 3,13-cyclized cembrane skeleton, named flaccidane.
The cytotoxicity of compounds 1–11 against the HT-29 (Human colon adenocarcinoma cell), P388 (mouse lymphocytic leukemia cell line), and K562 (Human erythromyeloblastoid leukemia cell line) tumor cell lines were determined. The results showed that compounds 7 and 10 were found to exhibit moderate cytotoxicity toward HT-29, P388 and K562 cells (Compound 7 IC50 = 15.09, 14.81 and 12.69μM; compound 10 IC50 = 13.18, 18.91 and 9.71μM). Compounds 7, 9 and 10 were found to show activity against superoxide anion and elastase release. Especially, compound 9 was found to show significant activity, and IC50 were 0.34 ± 0.01 and 0.35 ± 0.04 μM
