Study of the protective effect of Sargassum hemiphyllum extracts on influenza type A virus (H1N1) infection / 探討半葉馬尾藻萃取物對於 A 型流感病毒 (H1N1) 感染之保護效果

Hence, there is an urgent need for new treatments against these emerging viral infections. Influenza is an acute respiratory illness caused by infection of influenza virus. The outbreaks are very common throughout the world resulting in significant morbidity and mortality particularly in high risk patients. Recently, the extracts of algae have been shown broad biological activities, including antivirus, antitumor, immunomodulation and anticoagulant activity. In this thesis, we studied the effects of brown seaweed (Sargassum hemiphyllum) extracts on influenza A virus (H1N1) infection, where in vitro and in vivo model, were used plaque assay, viral RNA expression (real-time PCR), and immunofluorescence assay to study the protective effects of Sargassum hemiphyllum extracts. The inhibition effect of Sargassum hemiphyllum extracts on influenza virus infection is at the adsorption stage. The Sargassum hemiphyllum extracts also have good antioxidant effects. In the infected BALB/c mice model, 6 weeks-old mice were injected intranasally with lethal doses of influenza virus, on which the mortality rate was 100%. When the mice pre-treated with Sargassum hemiphyllum extracts before virus infection, the mortality rate was have effectively reduced. The Sargassum hemiphyllum extracts also as anti-inflammatory agents effectively decrease the lung injury in the influenza A virus (H1N1)-infected mice. In summary, the Sargassum hemiphyllum extracts show a protective effect against influenza virus infection on both cell and animal experiments. Sargassum hemiphyllum extract may be an ideal material to develop into anti-influenza virus agents.
