Preparation and analysis of alginate/cyanophycin composite / 海藻酸接枝藍藻蛋白之製備與鑑定

碩士 / 國立臺灣科技大學 / 化學工程系 / 103 / Compare with the natural and recombinant Cyanophycin Granules Protein(CGP), both types of CGP consists of Aspartic acid(Asp) and Arginine(Arg), but the recombinant CGP has Lysine(Lys) structure which contain primary amine and can be widely used for modification of proteins.

CGP is a natural and biocompatibility protein. However, because of its weak mechanical strength and low solubility, the CGP doesn't be commonly used for biomaterial. In this study, using alginate, which has good biocompatibility and biodegradability, to graft on different molar percentage of soluble CGP. Through the TNBSA method, the group, expected more quantity of CGP, detected more CGP experimentally. Due to the special structure of alginate, it can easily merge with divalent cations to become gels and now is widely used in drug encapsulation. After experimenting with different concentration of calcium chloride, 2M CaCl2 has been chosen for the crosslinking agent. Different percentage groups of samples are Freeze-dryed, and then cross link with 2M CaCl2. Finally we can get the 3-D structure called scaffold. All of the scaffolds can rapidly and sufficiently absorb moisture via swelling ratio test. Besides, the results show that all groups have pores through the scanning electron microscope (SEM).The more quantities of alginate in scaffold, the bigger size of pores.

In conclusion, after the alginate successfully graft on soluble CGP, it can become a scaffold which has pores and can absorb moisture. Observe whether using the scaffold for culturing can promote a proliferation of cells like chondrocyte or not in the future.
