The Effect of Antarctic Krill Oil to Learning and Memory in Senescence Accelerated Mice / 探討南極磷蝦油改善老化促進小鼠學習記憶之研究

碩士 / 靜宜大學 / 食品營養學系 / 103 / The decline of learning and memory with aging and AD due to the increased inflam-matory response and lower neurotrophic factor. Antarctic krill oil contains abundant omega-3 polyunsaturated fatty acid , astaxanthin and antioxidant. Antarctic krill oil was demonstrated to have many health beneficial effect, such as antidepressant and anti-inflammation. However, the effects of Antarctic krill oil on learning and memory is still unclear. This study aimed to examine the effect of Antarctic krill oil on learning and memory in senescence Accelerated mice. Six-month old SAMR1 mice was used to be normal control, while SAMP8 mice were divided into control, 2%fish oil、0.5%、1% and 2% Antarctic krill oil groups. In 12 weeks experiment period, food intake, body weight, single-trial passive avoidance test and active shuttle avoidance test were performed. After sacrificed, we evaluated brain β amyloid and inflammatory factors such as COX-2, iNOS, TNF-α and EP3 of brain, cortex, hippocampus and striatum were analyzed. Moreover, we also evaluated the and neurotropic factor, such as brain-derived neurotrophic factor (BDNF), TrkB receptor and cAMP response ele-ment-binding protein (CREB) mRNA expressions. The results showed. SAMR1 and SAMP8 fed fish oil or Antarctic krill oil diets had Retard the degradation of learning and memory, and were similar with SAMR1 group. and significantly decreased β am-yloid in brain Antarctic krill oil administration significantly decreased COX-2, iNOS, TNF-α in cortex, hippocampus and striatum and EP3 in the hippocampus expressions and lower than fish oil. Antarctic krill oil also increased BDNF, CREB expressions in cortex, hippocampus and striatum, and higher than fish oil whereas the TrkB receptor expression did not differ. In both of neurotrophic factors and inflammatory factors have shown a dose-dependent effect. In summary the supplementation of Antarctic krill oil could reduce inflammatory response, increase neurotrophic factor, then Im-prove learning and memory.
