The Effect of Emerging Contaminants on Neocardina denticulate Genome under Laboratory Conditions / 實驗室條件下多種新興汙染物對 多齒新米蝦(Neocaridina denticulate) 的基因表現影響

碩士 / 東吳大學 / 微生物學系 / 103 / Emerging pollutants, according to Environmental Protection Administration, is defined as newly recognized or not recognized chemical pollutants. Most of them are Persistent Organic Pollutants (POPs), such pollutants usually emerged via human activities. With the characteristics of difficult to decompose, they are easy to accumulate in the environment. Previous studies tend to study these pollutants with single pollutant instead multiple pollutants. By using Expressed Sequence tags(ESTs) of Neocaridina denticulate, effects of pollutant were evaluated with viability and gene expression. However, in the environment, those pollutants do not accumulate singly. To establish a better way of evaluation, we used three emerging pollutants in this study. They are (1) Pharmaceuticals and Personal Care Products as Pollutants (PPCPs), (2) Endocrine Disrupting Chemicals (EDCs), (3) industrial chemicals (such as plasticizing agents and brominated flame retardants). Gene expression and physiological activities were examined after various days of exposure to the pollutants.Our current effluent standard emphasis on the chemical method, it is difficult to effectively control emerging contaminants, and a wide variety of chemical substances difficult exile standards set for all single compound, so the use of biological detection, water toxicity as standard, will be the future development direction.
In this study, the expressed sequence tag (ESTs) system developed from shrimp Neocaridina denticulate, were use to detection of four group aquatic toxicity of pollutants, including Nonylphenol(NP), four Phthalate esters (PAEs), two Brominated flame retardant (BFRs), and two Personal Care Products (PPCPs). In the mixing of different drugs, PPCPs group with different concentrations ACE / IBU were mixed, with the proportional increase in concentration (0.1 / 0.1,0.3 / 0.3,0.5 / 0.5 mg / L) respiration and metabolism-related gene expression from inhibition become a trend promoted, and defense-related genes are rising trend, Test time stretched to three days, the upward trend will appear in the lower concentration (0.3 / 0.3 mg / L), it shows the pressure caused by the drug will accumulate over time, Other groups such as nonylphenol alone (0.5 mg / L) two kinds of BFRs (TBBPA 0.025 ppm, DBDE 0.025 ppm) four kinds of PAEs (DEP 0.1 ppm, DPP 0.1 ppm, DPrP 0.1 ppm, DHP 0.001 ppm), Test 1 , five days, most of gene expression have significant differences, but there is no clear trend. Where defense-related genes 70 kD heat shock-like protein is particularly noteworthy, soaked in the drug test either alone or in combination of performance, mostly the gene expression increased markedly, but in this experiment, mixing different categories of environmental hormones but are suppressed the expression of this gene.
