Studies on the Chemical Constituents and Anti-inflammatory Activities From the Stem of Pittosporum illicioides var. illicioides / 疏果海桐莖部之化學成分及抗發炎抗發炎活性之研究

Pittosporum illicioides var. illicioides (Pittosporaceae) is an evergreen shrub distributed in medium to high altitude in forests of China and Taiwan. In our studies on the anti-inflammatory constituents of Formosan plant, many species had screened for in vitro anti-inflammatory activity, and P. illicioides var. illicioides was found to be one of the active species.
Six new phthalides, (E)-3-ethylidene-7-hydroxy-6-methoxyphthalide (1), (E)-3-ethylidene-7-hydroxy-5,6-dimethoxyphthalide (2), (E)-3-ethyli- dene-5,6,7-trimethoxyphthalide (3), (S)-3-ethyl-5,7-dihydroxy-6- methoxyphthalide (4), (R)-3-ethyl-7-hydroxy-5,6-dimethoxyphthalide (5), (R)-3-ethyl-4,7-dimethoxyphthalide (6), and 16 known compounds (7–22) were isolated and identified from the n-hexane- and EtOAc-soluble fractions of stem of P. illicioides var. illicioides. Sixteen known compounds include four phthalides, (S)-3-ethyl-7-hydroxy-5,6-dimethoxy phthalide (7), (S)-3-ethyl-7-hydroxy-6-methoxyphthalide (8), (R)-3-ethyl- 7-hydroxy-6-methoxyphthalide (9), (Z)-3-ethylidene-6,7-dimethoxy- phthalide (10), two coumarins, aesculetin dimethyl ether (11), 6,7,8- trimethoxycoumarin (12), five benzenoids, vanillic acid (13), syringic acid (14), syringaldehyde (15), 2,4-dihydroxy-6-methoxyacetophenone (16), sinapaldehyde (17), a benzoquinone, 2,6-dimethoxy-1,4-benzoqui- none (18), two lignans, (-)-syringaresinol (19), glaberide I (20), and two steroids, mixture of β-sitostenone (21) and stigmasta-4,22-dien-3-one (22). The structures of all isolates were determined through spectral analysis and comparison of their physical and spectral data with literatures.
Among the isolated compounds, (E)-3-ethylidene-7-hydroxy-6- methoxyphthalide (1), (E)-3-ethylidene-7-hydroxy-5,6-dimethoxy phthalide (2), (E)-3-ethylidene-5,6,7-trimethoxyphthalide (3), (S)-3-ethyl- 5,7-dihydroxy-6-methoxyphthalide (4), (R)-3-ethyl-7-hydroxy-5,6- dimethoxyphthalide (5), (R)-3-ethyl-4,7-dimethoxyphthalide (6), (S)-3- ethyl-7-hydroxy-5,6-dimethoxyphthalide (7), (Z)-3-ethylidene-6,7- dimethoxyphthalide (10) exhibitited inhibition (with IC50 values in the range of 0.64~6.66 g/mL ) of superoxide anion generation by human neutrophils in response to formy-L-methionyleu-L-lckyl-L-phenyla- lanine/cytochala-sin B (fMLP/CB). (E)-3-Ethylidene-7-hydroxy-6- methoxyphthalide (1), (E)-3-ethylidene-7-hydroxy-5,6-dimethoxy (2), (E) -3-ethylidene-5,6,7-trimethoxyphthalide (3), (S)-3-ethyl-5,7-dihydroxy- 6-methoxyphthalide (4), (R)-3-ethy-7-hydroxy-5,6-dimethoxyphthalide (5) inhibited fMLP/CB-induced elastase release with IC50 values in range of 1.02~4.96 g/mL.
