Functional analyses of sea urchin PRL in mesenchymal cells during embryonic development / 胚胎發育時期海膽PRL於間葉細胞的功能分析

碩士 / 慈濟大學 / 分子生物暨人類遺傳學系碩士班 / 103 / Phosphatase of regenerating liver, PRL, is a member of PTP superfamily and has been identified as an oncogene. PRL regulates cell proliferation, differentiation, and involves in metastasis of various cancers. The vertebrate PRL family members comprise PRL-1, PRL-2 and PRL-3, while there is only one PRL in invertebrates. The amino acid sequences of PRLs are highly conserved among species. The sea urchin Strongylocentrotus purparatus PRL (SpPRL) shares 78% sequence similarity with human PRL-1. In this study, I used sea urchins as an animal model to study the role of PRL during embryogenesis. The SpPRL was highly enriched in secondary mesenchymal cells that are capable to migrate during early embryogenesis. Later, SpPRL was also found to be expressed in skeletogenic cells, pigment cells, and cells facing the lumen of archenteron. The suppression of SpPRL activity with PRL inibitors reduced the numbers of mesenchymal cells including the primary mesenchyme cells, pigment cells, and the precursor of pigment cells. In addition, the skeletal rods of skeletogenic cells and pseudopodia of pigment cells were malformed. I also showed that the inhibition of SpPRL enzymatic activity caused apoptosis in mesenchyme cells. As the supression of apoptosis did not fully rescue the developmental defects of spicule formation and pesudopod formation, we conclude that SpPRL is also involved in the skeletogenesis and the formation of pesudopod in addtion to its function in the regulation of cell viability. This study not only expands our understanding of PRL in embryogenesis, but also provides insights for understanding the mechanism of PRL-related metastasis.
