Regulation of hepatic lipid transportation by high fat treatment in vitro and in vivo / 探討高脂在in vitro及in vivo對肝臟脂質運輸的調控

碩士 / 國立陽明大學 / 生化暨分子生物研究所 / 103 / Non-alcoholic fatty liver disease (NAFLD) is a progressive liver disease worldwide. Both obesity and dyslipidemia play important roles in the NAFLD progression. To examine how obesity influence the progress of NAFLD pathologies, we stablished an obese animal model to study the effect of high fat diet (HFD) on hepatic lipid accumulation and the regulation of lipid transportation protein expression. We found that the levels of body weight, blood glucose, fatty liver infiltration, hepatic total cholesterol, serum total cholesterol and hepatic triglycerides were all increased and the expressions of hepatic lipid transportation proteins ATP-binding-cassette transporter A1 (ABCA1), low density lipoprotein receptor (LDLR) and low density lipoprotein receptor related protein (LRP) were all down-regulated in C57BL/6 mice fed with 60% HFD. Chinese olive extract (COE) contains polyphenols which may have the potential in alteration of hepatic lipid metabolism and body weight. In the present study, we investigated the effect of COE on the physiological progress of fatty liver and the HFD-induced obesity. From the results, COE supplementation not only mildly decreased body weight, serum cholesterol and fatty liver infiltration, but also increased the protein expression of hepatic LDLR compared with the 60% fed HFD mice. The regulatory mechanisms of lipid transportation were also evaluated in Huh7 cells treated with high doses of fatty acids in the presence of COE. It seems that COE is able to increase the protein expression of LDLR in Huh7 cells. Taken together, COE not only reduces body weight and prevents hepatic lipid accumulation in vivo but also increases the LDLR protein expression in vitro, suggesting its promising potential for the prevention of NAFLD progression.
