碩士 / 國立高雄海洋科技大學 / 海洋生物技術研究所 / 104 / Acute hepatopancreatic necrosis disease (AHPND) has caused a huge economic loss in shrimp industry in recent years after its outbreak in China in 2009. The pathogen is a specific strain of Vibrio parahaemolyticus (VPAHPND) which can cause up to 100% mortality of shrimp larval within 30 days after their stocking to the ponds. This study is to develop a rapid, accurate diagno-sis of VPAHPND based on a Real-time PCR. The test will take 1.5 hours, and its detection of virulence gene can be as low as to one copy. The specific primers were designed so that only VPAHPND pathogen was identified as compared with 24 non-AHPND V. parahaemolyticus and 10 other aquaculture pathogens. According to our survey for the Litopenaeus vannamei of culture ponds, there were 30% of larval and 26% for the adult shrimp were carriers of VPAHPND. 64.29% water sample of shrimp ponds were diagnosed with VPAHPND positive. The kit developed for detection of VPAHPND with specific primers based on the SYBR green real-time PCR has been proved to be successful. In addition to screen the VPAHPND for larval, this kit can also be used to screen the pathogen contamination in shrimp feed.
