The signal transduction of dopamine modulate the immunological responses in Macrobrachium rosenbergii / 多巴胺調節淡水長臂大蝦免疫反應之訊息傳遞途徑

碩士 / 國立屏東科技大學 / 水產養殖系所 / 104 / Dopamine (DA) was found to influence the immunological responses and resistance to pathogen infection in invertebrates. To clarify the possible modulation of DA through dopamine receptor (DAR) against acute environmental stress, the levels of DA, glucose and lactate in the haemolymph of Macrobrachium rosenbergii under hypo- (15 °C) and hyperthermal (34 °C) stresses were measured. Prawn received different dopamine receptor antagonists, such as SCH23390 (D1 DAR antagonist), domperidone (D2 DAR antagonist) and chlorpromazine (D1+D2 DAR antagonist) followed by hypo- and hyperthermal stresses to detected the alteration of immune parameters such as total haemocyte count (THC), different haemocyte count (DHC), phenoloxidase (PO), respiratory bursts (RBs), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activity and phagocytic activity (PA). In addition, pharmacological analysis of the effect DA modulation was studied in haemocytes incubated with DA and DAR antagonists (in vitro); and to know more about the modulation of DA and DAR in prawns were used the same pharmacological method in injection treatment to detected the alteration of immune parameters (in vivo). The results showed when prawn was exposed with hypo- and hyperthermal stresses for 2 hours, dopamine, glucose and lactate of haemolymph significantly increased. The results suggest that the upregulation of DA, glucose and lactate in haemolymph might be the response to acute thermal stresses for the demand of energy. Meanwhile prawn was injected with DAR antagonists and exposed with thermal stresses for 30 minutes, only in hyaline cell and RBs found the significantly difference. In vitro experiment showed that dopamine through D1 DAR to regulated RBs and SOD activity, and through D1 and D2 DAR to regulated GPx activity, however in PO activity not showed significantly difference. In vivo experiment also showed that the D1 DAR regulated RBs activity, and D1 and D2 DAR regulated PA, however in vivo experiment for SOD and GPx not showed significantly difference. The conclusion is thermal stress induced stress response in prawn, the physiological responses were earlier than immunological responses. And dopamine can through D1 DAR to regulated THC and RBs, and through D1 and D2 DAR to regulated PA, however additional studies in vitro and in vivo are necessary to elucidate the difference results of SOD and GPx activity.
