碩士 / 國立清華大學 / 系統神經科學研究所 / 104 / Alzheimer’s disease (AD) and Parkinson’s disease (PD) are the two most prevalent neurodegeneration disorders, but our understanding to these diseases is limited. With only a few pathological-related genes being identified, many studies now focus on finding more genes by genome-wide association study (GWAS). In this study, we used Drosophila eye model to test AD and PD associated genes reported in some recent studies. By up- and down-regulating several genes in fly carrying human tau or amyloid-beta 42 (Aβ42), we verified experimentally the phenotypes of the eyes. Our results reveal that some of the genes reported to interact with Aβ42, including PSEN2, CLU, PICALM and PLD3, also mediate tau toxicity, and MEF2C gene shows modifications in Aβ42. PD associated genes identifying suggests the pathogenesis relating to lysosome dysfunction.
