A study of surfactin in anticancer and anti-aging / 海洋活性物質surfactin抗腫瘤及抗老化之研究

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 104 / The surfactin refer to a Bacillus subtilis amphiphilic molecular which consists of a cyclic lipopeptide. Surfactin not only has the surface tension but also can reduce stabilize emulsions as a potent type biosurfactant. Surfactin consists of a heptapeptide and C14–15 -hydroxy fatty acid. Studies indicate that surfactin have a variety of bioactivities including anti-bacterial, anti-viral, anti-inflammatory and anti-tumor. Submerge fermentation due to high moisture increases the bacteria contamination risk, in operation for an aseptic and defoaming agents will affect a purification process. Macromolecules of skin care products is hard to penetrate dermal layer, therefore to employ skin penetration enhance play an import role to achieve the effect. In recent years, cancer become the top cause of death in Taiwan. Currently many studies focus on the separation and purification of natural products, extract an effective anticancer substances, replace chemicals with natural products to inhibit cancer cell growth and initiation of apoptosis. The objectives of this study is to develop on new method to produce surfactin, applied in cosmetic and anti-tumor. To fulfill this objective, in present study, three short-range goals: 1)to establish production of surfactin by Bacillus subtilis use semi-solid state fermentation, and establishing activity measurement method, 2)to employ surfactin used as a the skin penetration enhancers investigate cosmetics regulator (Puerariae Radix) on the cosmeceutical products, 3)to determine the mechanisms of surfactin on anti-tumor cell.
Experimental results showed that the yield of using semi-solid state fermentation method to produce surfactin is about 5.5g / kg. Surface tension surfactin is from 68 mN/m down to 25 mN/m. The critical micelle concentration is about 0.01 mM. Surfactin used as transdermal penetration enhancer can enhance the skin the effect of Puerariae Radix anti-aging, whitening and cell proliferation in human skin fibroblast cells. The results of present study showed that surfactin can induce apoptosis and inhibit cell cycle. In present study, the surfactin is produced by semi-solid state fermentation and use soybean as substrate, soybean powder with containg surfactin can be used as nutraceuticals to inhibit tumor cell proliferation.
