博士 / 國立臺灣海洋大學 / 水產養殖學系 / 104 / The effects of fucoidan on the immune response of white shrimp Litopeneaus vannamei haemocytes were studied in vitro and in vivo. Shrimp received fucoidan induced degranulation that caused changes in the cell morphology, cell size, and reduction in cell viability, increased in phenoloxidase (PO) activity, and respiratory burst (RB) in vitro.
Shrimp received fucoidan showed haemocyte proliferation and a higher mitotic index of hematopoietic tissue (HPT). The immune parameters including hyaline cell (HC), granular cell (GC), total haemocyte count (THC), PO activity, RB, and superoxide dismutase activity (SOD) were examined when white shrimp were injected with fucoidan at 0, 2, 6, and 10 µg g-1. HC, GC, THC, RB, and SOD increased to a plateau after 48 h, whereas PO activity increased to plateau after 72 h and were decreased to the background value after 144 h. Phagocytosis and clearance efficiency against Vibrio alinolyticus were significantly higher in shrimp received fucoidan at 2, 6, and 10 µg g-1. Resistances of shrimp against V. alinolyticus and white spot syndrome virus (WSSV) were higher over 48-144 h in shrimp that received fucoidan at 48 h and 144 h post-challenge.
Immune parameters of white shrimp received fucoidan 0, 2, 6, and 10 µg g-1 then challenged with V. alinolyticus 6  106 cfu shrimp-1 for 48 h and 144 h were examined for immune parameter. The HC, GC decreased earlier than THC, PO, RB, and SOD activity at 48 h post challenge, whereas at 144 h post challenge, the immune parameter of shrimp that received V. alinolyticus were deceased earlier. Shrimp received fucoidan at 0, 2, 6, and 10 µg g-1 were challenged with WSSV 1.2 × 105 copies shrimp-1 for 48 h and 144 h post challenge. The immune parameters of shrimp received fucoidan were significantly higher than those of control shrimp. LGBP, proPO I, proPO II and haemocyte homeostasis-associated protein (HHAP) gene expressions significantly increased in shrimp that received fucoidan at 6 µg g-1 of 48 h.
Shrimp immersed in seawater containing fucoidan at 0 (control) and 200 mg L-1 for 3 h and then exposed to 5 mg L-1 ammonia-N, were examined for immune parameters 6-24 h post-stress. The GCs, PO activity and RB of shrimp immersed in seawater containing fucoidan at 200 mg l-1 returned to original values at 12-24 h post-stress. The HCs and THC of shrimp immersed in seawater containing 200 mg L-1 fucoidan were significantly higher than those of control shrimp at 12-24 h post-stress.
Shrimp fed diets containing 0, 0.5, 1.0, and 2.0 g kg-1 fucoidan for 21 days were challenged with V. alginolyticus at 106 cfu shrimp-1. Phagocytic activity and clearance efficiency of white shrimp against V. alginolyticus were significantly higher in shrimp fed 1.0 g kg-1 diet compared to those of shrimp fed 0, 0.5, and 2.0 g kg-1 diets.
In conclusion, fucoidan activates immunity in vitro, and increases mitotic index of HPT, proliferation, immune parameter, LGBP, proPO I, proPO II, astakine and HHAP gene expression as well as resistance against pathogens V. alginolyticus and WSSV infection. Shrimp received fucoidan increased resistance against V. alginolyticus and WSSV after 48 h and144 h and showed high performance in immune parameter after challenged with V. alginolyticus and WSSV. Dietary administration of fucoidan at 1.0 g kg-1 enhanced the immune response of shrimp and resistance against V. alginolyticus infection. Shrimp that received fucoidan and subjected to ammonia stress showed a capability for positive regulation by an earlier recovery of immune parameters in maintaining homeostasis.
