Protective efficiency of Bioencapsulated oral bacterins of formalinzed and sonicated Vibrio parahaemolyticus and V. vulnificus in cultured grouper / 以生物殼材包覆福馬林不活化及超音波震碎腸炎及創傷弧菌作為口服疫苗對養殖石斑之保護效果

碩士 / 國立臺灣海洋大學 / 水產養殖學系 / 104 / This study investigated the enhancemeat of Vibrio antigen presentation releasing to promote protective efficacies of oral vaccines in cultured grouper. Vibrio cells were treated with three ways, formalin inactivated, ultrasound sonicated and frozen. Higher ELISA absorbance(ABS) represented higher amount of antigen presentation. Formalin could inactivate pathogen by denaturing its proteins, but result in the decrease of antigen releasing amount. Frozen treatment did not affect the amount of antigen presentation, and the ultrasound sonication shatter treatment took too long for completely killing all Vibrios. Therefore, frozen and ultrasound sonication were combined to inactivated the bacteria. After ultrasound sonication, Vibrios were frozen for completely killing.
Antigens releasing amount in each treatment was obtained in this study. In sodium alginate encapsulated group, raising the concentration of sodium alginate and calcium chloride to 2.5% could release the highest antigen amount. In cellulose acetate phthalate (CAP) encapsulated group, the use of 5% CAP could result in the highest amount of antigen releasing.
Groupers were i.p. challenged with 5 times LD50 of Vibrio. parahaemolyticus and V. vulnificus respectively after two times of oral vaccination with sodium alginate encapsulated bacterins and cellulose acetate phthalate encapsulated bacterins. In 10 times LD50 challenge test, formalin’s group reached highest RPS at 8 wpv and sonicated group reached highest RPS at 12th wpv. In alg-sonicated group RPS could reach 100% while CAP-formalin’s group only could reach 50%. .
In serum antibody titer analysis, each treatment and control groups had significant differences (p <0.05). Values of serum antibody titer increased significantly at 4th wpv, then reached highest at 12th wpv and then decreased and remained stable. Values of sonicated group were higher than that of formalin’s group from start to 20th wpv.
In Superoxide dismutase(SOD) analysis test, the SOD value reached the highest at 4th wpv then decreased. Values of alginate sonicated group was higher thanother group.
After vaccination, alginate-sonicated group exhibited the best performance in RPS, serum antibody titers and SOD value , followed by CAP - sonicated group, alginate – formalin’s group, the worst is CAP – formalin’s group. The protective efficacies in duration of CAP – ultrasound sonicated group was better than that of other groups
