The microfluidic chip device can makesurements the surrounding oxygen consumption (production) rate, for understand the impact of health chlorella, from different concentrations of herbicide (Diuron) . Herbicide (Diuron) ,for inhibitors the photosynthetic electron transport chain , it will not affect the role of mitochondrial respiration by different impact of the herbicide (Diuron). In accordance with the change in the oxygen generation rate of Chlorella, it can determine the solution of herbicide (Diuron) amount of concentration to achieve the purpose of detection. The microfluidic drvice has two components: PMMA microwell array chip , it include Pt(II) octaethylporphyrin the layer for Oxygen senser、colloidal graphite layer to blocking the light and Cu mesh layer coated with Agarose; Pneumatic closure device, can control to closed microchip that need to measure . Prevent the diffusion of oxygen in the amount of area measuring low oxygen measured is very important, Therefore, use of pneumatic closure device is closed microwell and use PMMA substrate denial culture liquid tank and the outside world to prevent the diffusion of oxygen to facilitate Chlorella low measurement of oxygen .
The experiment confirmed that the herbicide (Diuron) hardly affects mitochondrial respiration of chlorella, and chloroplast oxygenic photosynthesis rate in accordance with the herbicide (Diuron) concentration different from 0.2 μM to 1 μM and showed a linear decrease, the measurement for rate of change of the amount of oxygen generated results 0.2μM to 1μM between up to 55%, it is therefore demonstrated that the fabricated electrochemical biosensor can be an efficient indicator of water pollution.
