Inspection of Synthesis of Unsaturated Fatty Acids from Microalgae using Electrochemical technique / 以電化學模組分析探討藻類合成多元不飽和脂肪酸之代謝關係

碩士 / 元智大學 / 化學工程與材料科學學系 / 104 / Microalgae were a very important material for emerging green technology in recent year. It did not use methods by burning、chemical compounds and something will do damage to environment of human habitation. On the other hand, it even could produce high value added products by utilizing waste compound as nutrient source. In summary, microalgae offered a concept of sustainable development. In the presented study, the microalgae, recombinant Aurantiochytrium limacinum SR21, harboring vitreoscilla hemoglobin (VGB) gene was introduced to produce DHA at fermentation scale. The results showed concentration of biomass, content of total lipid and docosahexaenoic acid (DHA) were 18 gL-1, 11.95 gL-1 and 3.72 gL-1, respectively. Moreover, the recombinant microalgae displayed excellent performance in respect of higher lipid content under oxygen limiting cultivation (Total lipid 6.4 gL-1, DHA 1.82 gL-1) as compared to wild type strain.
As the consequence of it, the current (AC) impedance technique was subsequently conducted to investigate the relationship between electrochemical and biochemical performance of DHA fermentation. The electrochemical analysis depicted that ohm impedance and anode capacitor had high correlation with microalgae biomass and residual glycerol. The coefficient of determination (R2) is 0.8~0.85 and 0.85~0.9. It demonstrated that AC impedance could be a potential tool for establishing of DHA production model.
