The effect of the interaction between MBD2 and stress on adult hippocampal neurogenesis / MBD2和壓力的交互作用對海馬迴腦神經新生的影響

碩士 / 長庚大學 / 生物醫學研究所 / 105 / It has previously been suggested there is a strong link between hippocampal neurogenesis and emotional response. In this study, MBD2, a methylated DNA binding protein, was used to be involved in adult neurogenesis after unpredicted chronic stress. The aims of this study were (1) to understand the impact of the interactions between MBD2 and stress on the hippocampal neurogeneses of the mice, (2) to understand the impact of MBD2 on the responses of the mice to the stress and (3) to investigate the role of MBD2 in the mechanisms of hippocampal functions and neurogeneses, and its impacts on behaviors of the mice. The materials for experiments were MBD2 knockout and wild-type mice. The experimental items were mRNA quantification, behavioral tests (before and after stress introduction) and tests for the responses to the stress; immunochemical assays were applied to identify the hippocampal neurogeneses; the data were compared through the application of the independent t test to know if any difference between these two groups of mice really existed. The results revealed that the anxiety and depression-like behaviors of the two groups of mice were not significantly different before and after the stress induction; the baseline neurogeneses of the two groups of mice were not significantly different, but MBD2 knockout mice had presented a lowered level of neurogenesis; the plasma corticosterone level of the two groups of mice at baseline and 90 minutes after stress were significantly different. The mRNA expressions of glucocorticoid receptor, mineralocorticoid receptor, FKBP5 gene and serum and SGK1 gene of the two groups of mice were not significantly different before and after the stress introduction. To summarize those mentioned above, MBD2, the stress and the hippocampal neurogenesis should associate with each other in a specific way; however, further studies are needed to investigate the relationships among these three ones.
