碩士 / 中臺科技大學 / 醫學影像暨放射科學系暨研究所 / 106 / In the large scale production of microalgae biodiesel, high biomass productivity and high the lipid content of microalgae is the key. In order to obtain such microalgae, the microalgae of FGA326S (Secnedesmus) isolated from our laboratory was induced by ultraviolet radiation and cultured in a two-stage manner, and the lipids distribution of microalgae was observed by Nile red fluorescence staining technique and microscope equipment, and the lipid content was extracted simultaneously, to analyze and compare the lipid ratio (multiples) and the lipid content (percentage), in order to establish the optimum conditions for ultraviolet radiation treatment, and to obtain the FGA326S UV-treated strains to isolate the microalgae strains by ultraviolet radiation treatment, and finally to observe the cell phenotype of FGA326S wild type and UV-treated strains, and the growth changes and the lipid content of FGA326S wild type and UV-treated strains were analyzed. The results of this study showed that the lipid ratio (multiples) and the lipid content (percentage) of FGA326S microalgae change biggest when irradiated with UV-A+UV-B for 30 minutes. Mutagenesis and culture of FGA326S microalgae were carried out under this condition, and 6 strains the best growth of microalgae UV-treated strains, respectively, named M1~M6, comparison of wild type and mutated isolated strains M1~M6 lipid ratio (multiples), that M1 and M5 have the characteristics of high the lipid content, is a UV-treated strain of microalgae that has a great potential for producing lipid, and subsequent analysis of relevant properties to develop its use as an energy microalgae.
