Study of the photosynthetic pigments contents and antioxidant system in Scenedesmus sp. / 柵藻光合作用色素含量分析與抗氧化系統之探討

碩士 / 銘傳大學 / 生物科技學系碩士班 / 105 / Scenedesmus quadricauda was used as the control, and was compared with the photosynthetic pigment content and antioxidant system with Scenedesmus sp. MCU003 and Scenedesmus sp. MCU010, which was isolated at the Ming-chuan University, Taoyuan campus. S. sp. MCU003, S. sp. MCU010 and S. quadricauda data were show in the abstract. Data were analyzed using ANOVA test, P < 0.05.
We freeze-dried the cells when the cells were in the stationary phase. The biomass was 0.76 ± 0.17, 0.66 ± 0.03 and 0.65 ± 0.02 g D.W. / L respectively, and did not have a significant difference between each other. The Chlorophyll a content was 8.42 ± 1.15, 8.31 ± 1.21 and 2.75 ± 0.45 mg / g DW, the chlorophyll b contetnts were 2.11 ± 0.4, 2.12 ± 0.18 and 0.98 ± 0.02 mg / g DW, and the carotenoids contents were 3.33 ± 0.86,3.37 ± 0.58 and 1.52 ± 0.3 mg / g DW respectively. The contents of chlorophyll a, b and carotenoids, the experimental group were higher than the control group and reached the significant difference.
When cells reached the latter stage of the log phase, we collected cells and extracted by methanol after freeze-drying, the DPPH scavenging ability was 0.023 ± 0.008, 0.016 ± 0.002 and 0.026 ± 0.005 mg ascorbate / g DW, S. sp. MCU003 had a significant difference with the control group, but S. sp. MCU010 had no significant difference with the control. The phenolic contents were 17.99 ± 2.62, 13.5 ± 1.88 and 14.12 ± 1.63 mg gallic acid / g DW respectively, and had no significant difference with each other.
When cells reached the latter stage of the log phase, we broke live cells and collected to analysis the antioxidant system, CAT activity was 2.15 ± 0.3, 1.96 ± 0.18 and 2.4 ± 0.03 U / g; APX activity was 18.14 ± 0.67, 22.96 ± 0.48 and 39.34 ± 2.678 U / g protein respectively, the experimental group was lower than the control group, and also had a significant difference. The DPPH free radical scavenging ability was 0.015 ± 0.001 , 0.015 ± 0.0004 and 0.011 ± 0.001 mg ascorbate acid / g protein, each experimental group was higher than the control, and also had a significant difference. The polyphenol contents were 22.76 ± 0.76, 21.95 ± 0.47 and 30.48 ± 0.03 mg gallic acid / g protein respectively, the experimental group was lower than the control group, and also had a significant difference.
The methanol extraction of freeze-dried cells was tend to the highest S. quadricauda in DPPH free radical scavenging ability, but not significant, the polyphenol contents was no difference between each other. The APX activity in live cells extraction were the highest, but DPPH free radical scavenging ability was the lowest in S. quadricauda,which may result in similar antioxidant capacity of S. quadricauda and S. sp. MCU003. There is no significant difference in the antioxidant capacity of S. quadricauda and S. sp. MCU003.
