Effect of fish oil on plasma lipids and skeletal muscle in rats fed a diet rich in high fat and cholesterol / 魚油對攝食高脂飲食大鼠之血脂質及骨骼肌影響

碩士 / 國立臺灣海洋大學 / 食品科學系 / 105 / The aim of this study is to investigate the effect of fish oil on plasma lipid and skeletal muscle in rats fed high fat diet. Male Sprague-Dawley (SD) rats divided into three groups: (1) normal diet group (ND), (2) high fat diet group (HF) (15% lard + 2% soy bean oil), (3) high fat diet with 5 % fish oil (HO) (10% lard + 2% soy bean + 5% fish oil). All rats were fed the experiment diets and drinking water ad libitum for 8 weeks. The results showed that fish oil can decrease plasma total cholesterol and triglyceride levels. In addition, the LDL-C and VLDL-C concentrations were reduced by fish oil treatment. In liver, fish oil can induce AMP-activated protein kinase (AMPK) phosphorylation expression and reduce liver lipid synthesis. Fish oil can decrease plasma angiopoietin-like 4 (Angptl4) and apolipoprotein C3 (APOC3) protein expressions, and increased hepatic apolipoprotein E (APOE) and microsomal triglyceride transfer protein (MTTP) protein expressions, indicating that it may induce an increase in VLDL secretion. In skeletal muscle, fish oil can not only increase the AMPK phosphorylation expression, but also increase the muscle peroxisome –proliferatora ctivated receptor γ coactivator 1 α (PGC-1α) and peroxisome proliferator activated receptors γ (PPAR γ) protein expressions. Moreover, fish oil may increase uncoupling proteins 1 (UCP 1) expression, suggesting that fish oil may to regulate muscle lipid metabolism and decrease free fatty acid accumulation. In conclusion, fish oil may reduce hepatic lipid accumulation and skeletal muscle by increasing AMPK phosphorylation. Moreover, fish oil can increase the secretion of VLDL from liver by enhancing liver MTTP and APOE protein expressions and decrease plasma triglyceride by reducing ANGPTL4 and APOC3 protein expressions.
