Immunomodulatory effect of recombinant Macrobrachium rosenbergii nodavirus B2 protein against white tail disease of giant freshwater prawn M. rosenbergii / 淡水長臂大蝦野田病毒重組蛋白B2在提昇蝦體免疫能力及抑制白尾病之研究

碩士 / 國立高雄大學 / 生命科學系碩士班 / 105 / Giant freshwater prawn (Macrobrachiumrosenbergii, de Man, 1879) is an important commercial species due to its importance as food supply as well as a valuable export product. Since invertebrates lack a true adaptive immune response, stimulating M. rosenbergii for protection against white tail disease (WTD) with recombinant Macrobrachium rosenbergii nodavirus B2 protein is a possible approach. In the first experiment, prawns were injected with recombinant B2 protein. Samples were collected at different day intervals (6, 12, 24, 72, 168, 336 and 504 h) to analyze the hematological and immunological parameters and to study the immune-related and HSP gene expressions with β actin as internal control in hemocytes, gill tissue and hepatopancreas by RT-PCR. The expression of immune related genes such as superoxide dismutase (SOD), peroxinectin,anti-lipopolysaccharides (ALPs), Heat shock protein 90, Cytochrome oxidase and Manganese superoxide dismutaseand has been measured in prawn injected with recombinant B2 protein. In the second experiment, post-larvae (PL) of M. rosenbergii were exposed to recombinant B2 protein by immersion for 24 h with a similar dose of recombinant B2 protein and animals were challenged by immersion with MrNV. The samples were collected at different day intervals, i.e. 1st day, 2nd day and 5th day from treated group and 1st day, 5th day and moribund samples from MrNV infected PL group to analyze the expression of immune-related gene of M. rosenbergii by RT-PCR. The obtained percentage survival of post-larvae treated with purified B2 protein and challenged with MrNV and was 54% on 10th day p.i., 70% survival was recorded in normal group on 10th day p.i. and 10% survival was recorded in MrNV infected group on 10th day p.i. This suggests that the prawn immune system is able to specifically recognize and react with protein. This study shows that stimulation of the immune system of prawn with recombinant B2 protein helps to control WTD in giant freshwater prawn, M. rosenbergii.
